Job Title: Scientist, Epigenomics

Work Location: Suffolk County, Massachusetts

Summary:

Seeking a scientist to lead efforts in controlling genome-wide epigenetic reprogramming during oogenesis, focusing on building analytical and biological toolkits to measure, benchmark, and control epigenetic factors for in vitro oogenesis and high developmental competence of resulting eggs.

Responsibilities:

  • Design and execute pooled screens of epigenetic perturbations using engineered human iPSCs.
  • Develop and refine long-term culture systems to drive cellular differentiation into oogonia and subsequent stages.
  • Utilize advanced tools to analyze epigenetic marks at the single-cell level in both in vitro organoid systems and in vivo samples.
  • Integrate and interpret data from multiple epigenetic modalities, connecting epigenetic states to transcriptional programs and cell identities.
  • Identify and address upstream molecular events responsible for epigenetic abnormalities.
  • Innovate synthetic-biology approaches, including inducible transcription-factor and epigenetic-modifier circuits, to enhance reprogramming protocols.
  • Benchmark and ensure the developmental competence of in vitro generated eggs through comprehensive epigenetic analysis.
  • Collaborate with multidisciplinary teams to advance the understanding and control of epigenetic reprogramming in oogenesis.

Qualifications:

  • Exceptional scientific talent, creativity, and rigor.
  • Extensive knowledge of epigenetics, particularly DNA methylation dynamics and reprogramming in germ cells, early embryos, or iPSC systems.
  • Experience with human iPSC or stem cell culture.
  • Hands-on expertise in synthetic biology and genome engineering, including CRISPR knock-in/knockout, reporter and inducible-expression construct design, and pooled genetic screens.
  • Proficiency with epigenetic profiling techniques such as bisulfite/EM-seq, ChIP-seq/CUT&RUN, ATAC-seq, and Hi-C.
  • Strong computational skills for epigenetic data analysis, including bulk and single-cell RNA-seq and integration with epigenomic datasets.
  • PhD in stem cell biology, epigenetics, molecular or developmental biology, synthetic biology, or a related field; or demonstrated innovative work in the field.
